KAPN FIELD GUIDE / AROMA & FLAVOUR
Beta-Caryophyllene
“Caryophyllene” commonly refers to β-caryophyllene, especially the plant-predominant (E) isomer. It is distinct from α-caryophyllene (α-humulene).
Where it occurs naturally
The primary receptor paper reports (E)-β-caryophyllene in essential oils of black pepper, cinnamon and oregano, and as a major constituent of cannabis essential oil. These occurrence reports do not mean each sample contains the same amount. Sources: Beta-caryophyllene is a dietary cannabinoid
Aroma & flavour
Sensory literature associates β-caryophyllene with woody and spicy descriptors, including pepper-like notes. Sources: “Pepper”: Different Spices, One Name—Analysis of Sensory and Biological Aspects; Beta-caryophyllene is a dietary cannabinoid
What has research actually studied?
Evidence for CB2 activity depends on the assay. Gertsch et al. reported (E)-β-caryophyllene binding to CB2 and functional signaling in cells; oral compound also reduced carrageenan-induced inflammation in mice, but not CB2-knockout mice. Later, a human-CB1/CB2-expressing cell study found no change in membrane potential or modulation of CP55,940/THC responses; a follow-up found at most weak CB2 binding and no functional terpene effects. Sources: Beta-caryophyllene is a dietary cannabinoid; Absence of Entourage: Terpenoids Commonly Found in Cannabis sativa Do Not Modulate the Functional Activity of Δ9-THC at Human CB1 and CB2 Receptors; Terpenoids From Cannabis Do Not Mediate an Entourage Effect by Acting at Cannabinoid Receptors
What the evidence does not show
These are receptor/cell assays and a mouse model; later assays did not reproduce the earlier functional result, so findings differ across assay systems. None demonstrates a clinical effect or establishes that cannabis containing β-caryophyllene activates CB2 in consumers.
What does its presence in cannabis mean?
A product listing β-caryophyllene does not establish receptor activity or a predictable product experience. Isolated-compound findings cannot be applied to a whole cannabis product.
